Investigator HDplex Kit (100)

For forensic, human-identity and paternity testing, this QIAGEN multiplex assay co-amplifies 12 autosomal short tandem repeat (STR) loci together with the gender-specific Amelogenin marker in a single 25 µl PCR reaction. The kit includes primer mix, reaction mix, DNA polymerase, control DNA, allelic ladder HDplex, DNA size standard 550 (BTO) and nuclease-free water. The 12 loci — D2S1360, D3S1744, D4S2366, D5S2500, D6S474, D7S1517, D8S1132, D10S2325, D12S391, D18S51, D21S2055 and SE33 (ACTBP2) — contain markers not featured in commonly used standards, supporting reliable differentiation of forensic samples from related individuals and delivering heightened discriminatory power in kinship, paternity, immigration and difficult casework.

Primers are labelled with the fluorescent dyes 6-FAM, BTG and BTY, and amplicons are sized against the BTO-labelled 550 size standard and the BT5 matrix during capillary electrophoresis. The kit is intended for DNA profiling of blood, buccal swabs and forensic stains, with a recommended DNA input of 0.35 ng under standard conditions. Internal validations demonstrated robust and balanced results across 0.2–1.0 ng of input and reliable results with <0.1 ng DNA.

The kit has been validated on the GeneAmp PCR System 9700 thermal cycler and is compatible with the ABI PRISM 310, ABI PRISM 3100/3100-Avant, Applied Biosystems 3130/3130xl and 3500/3500xL Genetic Analyzers. Data can be analysed with Investigator IDproof, Investigator IDproof Mixture, GeneMapper ID, GeneMapper ID-X or Genotyper Software using the matching HDplex Template Files. All components are stored at –20°C, with the primer mix and allelic ladder kept protected from light, and the kit meets ISO 18385 requirements.
