QIAGEN offers a 6-dye multiplex STR solution designed for human identification, enabling simultaneous amplification of all Chinese National database loci, CODIS-required markers, European standard markers, DYS391 and Amelogenin in a single reaction. The kit co-amplifies 25 loci, providing very high discrimination power (PI 3.7 x 10⁻²⁹) for forensic genetic applications and paternity testing.

The reaction volume is 25 µl, with a recommended DNA input of 0.5 ng under standard conditions. Internal validations demonstrated robust and balanced results with 0.2 to 2 ng DNA, and reliable results with <0.1 ng DNA. The assay uses FRM 3.0 fast-cycling chemistry to deliver reliable amplification in approximately 60 minutes, with enhanced robustness towards PCR inhibitors and improved master mix stability at 4 °C. Performance was tested against six inhibitors: calcium, ethanol, indigo carmine, hematin, humic acid and tannic acid. The kit is intended for analysis of blood, buccal swabs and forensic stains, and is manufactured in alignment with ISO 18385. Once opened, the kit should be stored at 2–8 °C for a maximum of 6 months.

Fluorescence labelling uses 6-FAM, BTG, BTY, BTR2, BTO and BTP with the BT6 matrix standard. Analysis is performed on Applied Biosystems 3500/3500xL Genetic Analyzers using GeneMapper ID, GeneMapper ID-X or Genotyper software. The kit includes two internal Quality Sensor controls (QS1 and QS2) amplified alongside the STR markers to indicate PCR efficiency and the presence of inhibitors, aiding decision making for further sample processing. The DNA Size Standard is purchased separately, with options including DNA Size Standard 450 (BTO) offering 13 fragments for high-concentration inputs, or DNA Size Standard 24plex (BTO) with 26 fragments for standard applications.
