Insulin/IGF-1 signaling is a key barrier to the directed differentiation of pluripotent stem cells, and this 1 mg vial of IDE2 from STEMCELL Technologies provides a selective small-molecule route to attenuating that pathway. By inhibiting insulin receptor (IR) and insulin-like growth factor 1 receptor (IGF-1R), IDE2 enables precise temporal control during in vitro differentiation, supporting the generation of definitive endoderm, pancreatic beta-like cells, intestinal organoids, and hepatocyte-like lineages from human pluripotent stem cells.

The lyophilized powder is typically reconstituted in DMSO to a 10 mM stock and added directly to differentiation medium at working concentrations of 1–5 µM. IDE2 is widely cited as a substitute for exogenous insulin in stages where downregulation of pro-proliferative Akt signaling is desired, and has been validated in monolayer and embryoid body protocols employing cytokines such as activin A and FGF2.

The reagent is supplied for research use only and is not intended for diagnostic or therapeutic applications. Store the lyophilized material at -20 °C, protected from light; once reconstituted, aliquot and store at -20 °C to minimize freeze-thaw degradation.
