Description

STEMCELL Technologies Human/Mouse Recombinant NT-3 is a neurotrophic factor belonging to the nerve growth factor (NGF) family of proteins, which also includes NGF, brain-derived neurotrophic factor (BDNF), and neurotrophin-4/5. NT-3 signals through the receptor tyrosine kinase TrkC and promotes the survival, development, and differentiation of neurons, while also modulating transmitter release at synapses in the peripheral and central nervous systems.

The recombinant protein is expressed in E. coli and supplied lyophilized from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid. It is predicted to form a non-covalently linked homodimer with each dimer having a molecular mass of 13.8 kDa and a total predicted molecular weight of 27.5 kDa. Alternative names include HGNF, NGF-2, Nerve growth factor 2, Neurotrophin 3, and NT3 (UniProt accession P20783). For storage, the vial should be kept at -20°C to -80°C and is stable for 12 months from the date of receipt. Prior to use, the product should be reconstituted in sterile water to at least 0.1 mg/mL without vortexing.

Specifications include a purity of ≥ 95% and an endotoxin level of ≤ 1 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. Biological activity is demonstrated by proliferation of a neuroblastoma cell line stably expressing TrkB (BR6). For lot #1000050644 or higher, specific activity is ≥ 5.0 x 10^4 units/mg (EC50 ≤ 20 ng/mL); for other lots, specific activity is ≥ 50 units/mg (EC50 ≤ 20 μg/mL) using C6 cells, with a representative EC50 of 9.3 ng/mL in the bioassay example.
