Description

Vascular endothelial growth factor (VEGF-165) is a heparin-binding homodimeric glycoprotein that contributes to embryonic vasculogenesis and angiogenesis. The animal component-free reagent from STEMCELL Technologies binds to FLT1 (VEGFR-1) and KDR (VEGFR-2) and activates the Raf/MEK/ERK and PI3K/AKT signalling pathways. Multiple isoforms arise from alternative splicing of VEGF mRNA in the terminal exon, with proximal splice-site selection in exon 8 producing pro-angiogenic VEGFxxx isoforms and distal splice-site selection yielding anti-angiogenic VEGFxxxb isoforms. The factor also supports neurogenesis in vitro and in vivo, exerts neurotrophic effects on central nervous system neurons, and promotes the growth and survival of dopaminergic neurons, astrocytes, and vascular endothelial cells, as well as monocyte chemotaxis and colony formation by granulocyte-macrophage progenitor cells. Each VEGF-165 molecule is composed of two polypeptide chains of 165 amino acids.

The protein is expressed in E. coli and supplied lyophilised from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid. It has a predicted molecular mass of 19.3 kDa per monomer and 38.6 kDa as a homodimer, with a stated purity of ≥ 95%. The reported specific activity is ≥ 1.0 x 10^5 units/mg (EC50 ≤ 10 ng/mL), determined by a cell proliferation assay using human umbilical vein endothelial cells (HUVECs); in a representative assay, proliferation was measured after 88 hours of culture by a fluorometric method, yielding an EC50 of 3.35 ng/mL. Endotoxin is ≤ 1 EU/μg protein by kinetic Limulus amebocyte lysate (LAL) analysis. The reagent cross-reacts with mouse and rat samples and is associated with UniProt accession P15692-4. Vials should be stored at -20°C to -80°C and remain stable for 12 months from date of receipt. The manufacturer's quality management system is certified to ISO 13485, and the product is intended for research use only.
