Description

STEMCELL Technologies offers Human Recombinant TGF-beta 1, a member of the TGF-β superfamily that regulates diverse cellular phenotypes including cell proliferation, differentiation, wound healing, apoptosis, and metabolism. The protein binds to serine-threonine kinase type I and II receptors and activates signal transduction through SMAD2/3 proteins. During human embryogenesis, TGF-beta 1 is expressed by endothelial and hematopoietic tissues and acts as an endogenous autocrine growth regulator. Together with basic fibroblast growth factor (bFGF), it supports the culture of undifferentiated human embryonic stem cells and induced pluripotent stem cells. TGF-beta 1 is produced in latent form by many blood cells and, once activated, helps maintain immune homeostasis by modulating B cell response and mediating immunosuppressive effects on T cells and neutrophils.

Also known as cartilage-inducing factor, differentiation inhibiting factor, LAP (latency-associated peptide), TGFB1, and transforming growth factor β 1, this recombinant human protein is produced in CHO cells and carries UniProt accession number P01137. The predicted molecular mass is 12.8 kDa as a monomer and 25.6 kDa as a homodimer. Purity is ≥ 95% as assessed by SDS-PAGE. Bioactivity is determined by the ability to inhibit IL-4-induced HT-2 cell proliferation, with an EC50 of ≤ 0.5 ng/mL and a specific activity of approximately 2.151 x 10^4 IU/µg, calibrated against the human recombinant TGF-beta-1 WHO International Standard (NIBSC code: 89/514). Endotoxin level is ≤ 0.1 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis.

The product is supplied lyophilized from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid and trehalose. Store at -20°C to -80°C; the product is stable as supplied for 12 months from the date of receipt. To prepare, centrifuge the vial before opening and reconstitute in 10 mM hydrochloric acid to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. Repeated freeze-thaw cycles should be avoided, and stock solutions should not be stored at 2–8°C for more than 1 month or at -80°C for more than 3 months without testing for each application.
