Description

Sonic hedgehog (Shh) is a member of the Hedgehog family of secreted signaling proteins with a prominent role in patterning during early mammalian development. Recombinant human Shh (C24II; rhShh) from STEMCELL Technologies is a fully biologically active molecule. Shh is expressed during early embryogenesis in a variety of tissues and, during patterning of the central nervous system, directly acts on cells to specify neural cell fates, regulates proliferation and survival of oligodendrocyte precursors and neural crest cells, and stimulates proliferation, differentiation and axon growth. Shh also regulates T cell differentiation and activation.

The protein has a predicted molecular mass of 19.7 kDa and UniProt accession Q15465. Biological activity was tested by its ability to promote alkaline phosphatase production of CCL-226 cells, measured using a fluorometric assay method; the EC50 in the cited example is 0.7 μg/mL. The specific activity is ≥ 500 units/mg, corresponding to an EC50 ≤ 2 μg/mL, as determined by inducing alkaline phosphatase production of C3H/10T1/2 (CCL-226) cells. Purity is ≥ 95% as assessed by SDS-PAGE under reducing and non-reducing conditions with Coomassie Blue staining. The endotoxin level, measured by kinetic Limulus amebocyte lysate (LAL) analysis, is ≤ 0.2 EU/μg protein.

The product is expressed in E. coli and supplied lyophilized after dialysis against phosphate-buffered saline. It should be reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing. Store at -80°C, where the product is stable for 12 months from the date of receipt; avoid repeated freeze-thaw cycles, and limit storage at 2–8°C to no more than 2 weeks or at -20°C to no more than 3 months. Centrifuge the vial before opening. For research use only.
