Hu Recom Shh (C24II), 10 µg

STEMCELL Technologies supplies Human Recombinant Shh (C24II), also known as Sonic hedgehog (Shh), a member of the Hedgehog family of secreted signaling proteins that has a prominent role in patterning during early mammalian development. Shh is expressed during early embryogenesis in a variety of tissues. During patterning of the central nervous system, Shh directly acts on cells to specify neural cell fates, regulates proliferation and survival of oligodendrocyte precursors and neural crest cells, and stimulates proliferation, differentiation and axon growth. Shh also regulates T cell differentiation and activation.

The recombinant protein is produced in E. coli and has a predicted molecular mass of 19.7 kDa. It is formulated as a lyophilized product after dialysis against phosphate-buffered saline, with a purity of ≥ 95%. The endotoxin level is ≤ 0.2 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis.

Biological activity is determined by inducing alkaline phosphatase production of CCL-226 cells, with a specific activity of ≥ 500 units/mg (EC50 ≤ 2 μg/mL). In the cited example, the EC50 was 0.7 μg/mL. The product should be stored at -80 °C and is stable as supplied for 12 months from date of receipt. For reconstitution, the product should be dissolved in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing.
