STEMCELL Technologies supplies an animal component-free preparation of human recombinant RANKL (receptor activator of nuclear factor kappa-B ligand), a member of the tumor necrosis factor (TNF) superfamily of cytokines involved in long-term cellular activities such as differentiation, proliferation, and cell death. Also referenced by alternative names including CD254, TNFSF11, TRANCE, OPGL, and ODF, the protein is expressed as a type II homotrimeric transmembrane protein in both membrane-bound and secreted forms.

RANKL binds the receptor activator of nuclear factor kappa-B (RANK), activating the AKT signaling pathway. Osteoprotegerin (OPG) may also bind RANKL, competing with RANKL-RANK binding. The protein is involved in osteoclastogenesis and T cell activation. The product is lyophilized from a sterile-filtered solution containing sodium phosphate at pH 7.5, with a stated purity of ≥ 95%. Specific activity is ≥ 2 x 10^4 units/mg (EC50 ≤ 50 ng/mL), determined by the ability to induce bioluminescence in engineered RANKL bioassay cells, and the endotoxin level is ≤ 1 EU/μg protein measured by kinetic Limulus amebocyte lysate (LAL) analysis. According to the safety data sheet, the preparation is classified as not a hazardous substance or mixture under the Globally Harmonized System (GHS) and is supplied as a solid.
