Description

A recombinant form of the human Receptor activator of nuclear factor kappa-B ligand (RANKL), supplied by STEMCELL Technologies, this product belongs to the tumor necrosis factor (TNF) superfamily. The protein is a type II homotrimeric transmembrane molecule expressed in both membrane-bound and secreted forms. RANKL binds to the receptor activator of NF-κB (RANK) and, upon binding, activates the AKT signaling pathway. Osteoprotegerin (OPG) can also bind RANKL, competing with RANKL–RANK binding. The cytokine is involved in osteoclastogenesis and T cell activation.

Alternative names include CD254, hRANKL2, ODF, OPGL, OPTB2, Osteoclast differentiation factor, soluble Receptor activator of NF-κB ligand (sOdf), TNF-related activation-induced cytokine, TNFSF11, TNF superfamily member 11, TNLG6B, Tumor necrosis factor superfamily member 11, and TRANCE. The accession number is O14788, and the predicted molecular mass is 19.7 kDa. The protein is produced in E. coli and lyophilized from a sterile-filtered solution containing sodium phosphate at pH 7.5. Purity is ≥ 95%.

Specific activity is ≥ 2 x 10^4 units/mg (EC50 ≤ 50 ng/mL), as determined by the ability to induce bioluminescence in engineered RANKL bioassay cells. In an additional activity assay using RAW-Blue™ macrophage reporter cells with fluorometric detection of alkaline phosphatase, an EC50 of 38.8 ng/mL was measured. Endotoxin level is ≤ 1 EU/μg protein by kinetic Limulus amebocyte lysate (LAL) analysis. Store at -20 to -80°C; the product is stable as supplied for 12 months from the date of receipt. Centrifuge the vial before opening and reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. Optionally, dilute with concentrated bovine serum albumin (BSA) to a final BSA concentration of 0.1% after reconstitution. As a general guide, do not store at 2-8°C for more than 1 month or at -20 to -80°C for more than 3 months, and avoid repeated freeze-thaw cycles. For research use only.
