Description

STEMCELL Technologies offers Human Recombinant RANKL (Receptor Activator of Nuclear Factor Kappa-B Ligand), a member of the tumor necrosis factor (TNF) superfamily. The protein is expressed as a type II homotrimeric transmembrane protein in both a membrane-bound and a secreted (soluble) form. RANKL binds to the receptor activator of nuclear factor kappa-B (RANK), and upon binding it activates the AKT signaling pathway. Osteoprotegerin (OPG) can also bind RANKL, and this interaction competes with RANKL–RANK binding. The cytokine is involved in osteoclastogenesis and T cell activation, and TNF-superfamily members more broadly participate in long-term cellular activities such as differentiation, proliferation, and cell death.

The recombinant product is expressed in E. coli and has a predicted molecular mass of 19.7 kDa (UniProt accession O14788). It is supplied lyophilized from a sterile-filtered solution containing sodium phosphate at pH 7.5, with a reported purity of ≥ 95%. The endotoxin level is ≤ 1 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The specific activity is ≥ 2.0 x 10^4 units/mg, with an EC50 ≤ 50 ng/mL determined by embryonic alkaline phosphatase production induced in RAW-Blue™ macrophage reporter cells; a representative assay yielded an EC50 of 38.8 ng/mL.

For reconstitution, the vial should be centrifuged before opening and the product reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing; if not used immediately, it may optionally be diluted with concentrated bovine serum albumin (BSA) to a final BSA concentration of 0.1%. The lyophilized product should be stored at -20°C to -80°C and is stable as supplied for 12 months from the date of receipt. According to the manufacturer's Safety Data Sheet, the product is a laboratory reagent and is not classified as a hazardous substance or mixture under the Globally Harmonized System (GHS) or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200). It is intended for research use only and is not for human or animal diagnostic or therapeutic uses unless otherwise stated.
