STEMCELL Technologies offers a recombinant human cytokine, receptor activator of nuclear factor kappa-B ligand (RANKL), which is a member of the tumor necrosis factor (TNF) superfamily. RANKL is a type II homotrimeric transmembrane protein expressed in both membrane-bound and secreted form. It binds to the receptor activator of nuclear factor kappa-B (RANK); osteoprotegerin (OPG) may also bind RANKL, competing with RANKL–RANK binding. Upon binding to its receptor, RANKL activates the AKT signaling pathway and is involved in osteoclastogenesis and T cell activation.

The protein is produced in E. coli and supplied lyophilized from a sterile-filtered solution containing sodium phosphate at pH 7.5. It has a predicted molecular mass of 19.7 kDa and a purity of ≥ 95%, with endotoxin level ≤ 1 EU/μg protein. Specific activity is ≥ 2 x 10^4 units/mg (EC50 ≤ 50 ng/mL), as determined by the ability to induce bioluminescence in engineered RANKL bioassay cells. Alternative names include CD254, hRANKL2, ODF, OPGL, OPTB2, TRANCE, and TNFSF11.

Store at -20 to -80 °C; the product is stable as supplied for 12 months from the date of receipt. Before use, centrifuge the vial, then reconstitute by pipetting sterile water down the sides of the vial to a concentration of at least 0.1 mg/mL. Do not vortex. For research use only.
