Description

Manufactured by STEMCELL Technologies, this Human Recombinant R-Spondin-1 is expressed in CHO cells and supplied as a lyophilized powder from a sterile-filtered phosphate-buffered saline solution. The protein is also known by the alternative names hRspo1 and Roof plate-specific Spondin-1 (Accession Number Q2MKA7), with a predicted molecular mass of 26.8 kDa.

R-Spondin-1 is a member of the R-Spondin protein subfamily, characterized by a TSR-1 domain and two N-terminal furin-like cysteine-rich repeats. It acts as a potent enhancer of WNT signaling, activating β-catenin signaling via the WNT cascade and indirectly increasing low-density lipoprotein receptor-related protein 6 (LRP6) on the cell surface by binding leucine-rich repeat-containing G-protein-coupled receptor 5 (LGR5) and competing with the WNT antagonist DKK-1. The protein is required for the specification of hematopoietic stem cells and plays pleiotropic roles during embryogenesis; it has also been shown to be important in the growth, survival, and migration of ovarian cancer cells.

Product specifications include a specific activity of ≥ 2.0 x 10^4 units/mg (EC50 ≤ 50 ng/mL), as determined by luciferase activity induced in HEK-293 cells in the presence of mouse Wnt-3a, with a representative EC50 of 36.4 ng/mL. Purity is ≥ 95%, and the endotoxin level is ≤ 1 EU/µg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The product is stable for 12 months from the date of receipt when stored at -20 to -80°C, and should be reconstituted in sterile water to at least 0.1 mg/mL. It is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS) or the 2012 OSHA Hazard Communication Standard, and is not regulated for transport under DOT, IATA, IMDG, or other major transport regulations.
