Hu Recom PDGF-BB, ACF, 1000 µg

STEMCELL Technologies supplies Human Recombinant PDGF-BB in an animal component-free formulation. Platelet-derived growth factor (PDGF) is a dimeric glycoprotein composed of disulfide-stabilized polypeptide chains, assembled as homodimers (PDGF-AA, PDGF-BB) or heterodimers (PDGF-AB). PDGF signals through the receptor tyrosine kinases PDGFRα and PDGFRβ, and PDGF-induced migration has been shown to involve MEK/ERK, EGFR, Src, and PI3K/AKT signaling pathways. The factor functions as a mitogen for fibroblasts, glial cells, arterial smooth muscle cells, and epithelial and endothelial cells, and has been implicated in the pathogenesis of atherosclerosis, glomerulonephritis, cancer, and in the contraction of vascular smooth muscle cells.

PDGF-BB is secreted by osteoblasts to induce mesenchymal stem cell migration and angiogenesis, and is also secreted by preosteoclasts during bone modeling and remodeling to support proper osteogenesis. The product is expressed in E. coli and supplied as a lyophilized powder from a sterile-filtered aqueous solution containing sodium phosphate at pH 7.5. The predicted molecular mass is 12.4 kDa for the monomer and 24.9 kDa for the dimer, and purity is reported at ≥ 95% by SDS-PAGE. Endotoxin is measured at ≤ 1 EU/μg protein by kinetic LAL analysis. Cross-reactivity has been confirmed on human and rat cells.

Biological activity is determined by a cell proliferation assay using NR6R-3T3 cells, with an EC50 of ≤ 20 ng/mL (an example assay gave 5.41 ng/mL). The specific activity is approximately 1.66 x 10³ IU/µg, calibrated against the human recombinant PDGF-BB WHO International Standard (NIBSC code 94/728). The product should be reconstituted in sterile water to at least 0.1 mg/mL by pipetting without vortexing. It is stored at -20 °C to -80 °C and remains stable as supplied for 12 months from the date of receipt. The accession number is P01127, and the protein is also known by the alternative names GDGF (glioma-derived growth factor) and ODGF (osteosarcoma-derived growth factor).
