Description

STEMCELL Technologies supplies Human Recombinant PDGF-BB as a solid laboratory reagent. The product is not classified as a hazardous substance or mixture according to the Globally Harmonized System (GHS) or the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200), and 0% of the mixture consists of ingredient(s) of unknown toxicity. Recommended handling follows good industrial hygiene and safety practice, and storage should be in accordance with the Product Information Sheet.

The biological activity of Human Recombinant PDGF-BB was assessed by its ability to promote the proliferation of NR6R-3T3 cells, measured using a fluorometric assay method. The EC50, defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum, was 2.12 ng/mL. The protein is a homodimer of 12.4 kDa subunits with a predicted total molecular mass of 24.9 kDa, and 1 μg was resolved with SDS-PAGE under reducing and non-reducing conditions and visualized by Coomassie Blue staining.

Platelet-derived growth factor (PDGF) is a dimeric glycoprotein composed of two disulfide bridge stabilized polypeptide chains, A and B, which assemble as heterodimers (PDGF-AB) or homodimers (PDGF-AA and PDGF-BB). PDGF signals through the receptor tyrosine kinases PDGFRα and PDGFRβ, and PDGF-induced migration involves signaling pathways including MEK/ERK, EGFR, Src, and PI3K/AKT. PDGF is a mitogen for dermal fibroblasts, glial cells, arterial smooth muscle cells, vascular smooth muscle cells, epithelial cells, and endothelial cells, and it has been implicated in the pathogenesis of atherosclerosis, glomerulonephritis, and cancer, as well as in the contraction of vascular smooth muscle cells of rat aortic tissues. PDGF-BB is secreted by osteoblasts to induce mesenchymal stem cell migration and angiogenesis, and by preosteoclasts during bone modeling and remodeling to induce angiogenesis and proper osteogenesis. The product is not regulated for transport by DOT, TDG, MEX, ICAO (air), IATA, IMDG, RID, ADR, or ADN.
