Description

STEMCELL Technologies provides this recombinant human platelet-derived growth factor AB (PDGF-AB) for research use. PDGF is a dimeric glycoprotein consisting of two disulfide bridge-stabilized polypeptide chains, A and B, which assemble as heterodimers (PDGF-AB) or homodimers (PDGF-AA and PDGF-BB). The growth factor signals through the receptor tyrosine kinases PDGFR alpha and PDGFR beta, and PDGF-induced migration has been shown to involve signaling through MEK/ERK, EGFR, Src, and PI3K/AKT pathways. PDGF acts as a potent mitogen for cells of mesenchymal origin and has been implicated in processes including atherosclerosis, glomerulonephritis, cancer, and contraction of vascular smooth muscle cells.

The product is supplied as a lyophilized solid, prepared from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid, with E. coli as the expression source. Reported purity is ≥ 95%. Specific activity is ≥ 5 x 10^4 units/mg, with an EC50 ≤ 20 ng/mL as determined by a cell proliferation assay using BALB/c 3T3 cells. Endotoxin level is ≤ 1 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis. According to the safety data sheet, the product is not classified as a hazardous substance or mixture under the Globally Harmonized System (GHS).

Storage is recommended at -20°C to -80°C, and the product is stated to be stable as supplied for 12 months from date of receipt. Reconstitution is performed in sterile water to at least 0.1 mg/mL by pipetting down the sides of the vial without vortexing. Alternative names for the protein include GDGF, Glioma-derived growth factor, ODGF, and Osteosarcoma-derived growth factor. The product is for research use only and is not intended for human or animal diagnostic or therapeutic applications.
