STEMCELL Technologies offers this Human Recombinant PDGF-AB, a lyophilized recombinant growth factor expressed in E. coli. Platelet-derived growth factor (PDGF) is a dimeric glycoprotein composed of two disulfide bridge-stabilized polypeptide chains (A and B) that assemble as heterodimers (PDGF-AB) or homodimers (PDGF-AA and PDGF-BB). The protein signals through the receptor tyrosine kinases PDGFR alpha and PDGFR beta, and PDGF-induced migration has been shown to involve MEK/ERK, EGFR, Src, and PI3K/AKT signaling pathways. PDGF functions as a potent mitogen for cells of mesenchymal origin, including fibroblasts, glial cells, and vascular smooth muscle cells, and is also a mitogen for dermal fibroblasts, arterial smooth muscle cells, and epithelial and endothelial cells. The factor has been implicated in the pathogenesis of atherosclerosis, glomerulonephritis, and cancer, as well as in the contraction of vascular smooth muscle cells of rat aortic tissues. In combination with 5-Azacytidine, PDGF-AB has been shown to convert mature bone and fat cells into tissue-regenerative multipotent stem cells.

The product is supplied as a lyophilized powder from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid. Purity is ≥ 95%. Specific activity is ≥ 5 x 10^4 units/mg (EC50 ≤ 20 ng/mL), determined by a cell proliferation assay using BALB/c 3T3 cells, with an example EC50 of less than 1.8 ng/mL. Endotoxin level, measured by kinetic Limulus amebocyte lysate (LAL) analysis, is ≤ 1 EU/μg protein. The predicted molecular mass is 14.4 kDa (alpha monomer), 12.4 kDa (beta monomer), and 26.8 kDa (dimer). For use, the vial should be centrifuged before opening and the product reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. Store at -20°C to -80°C; the product is stable as supplied for 12 months from date of receipt.
