Hu Recom PDGF-AA, 100 µg

STEMCELL Technologies supplies this recombinant human platelet-derived growth factor as a lyophilized laboratory reagent for research use. The protein is a homodimer of two disulfide bridge-stabilized polypeptide chains, expressed in E. coli and purified to ≥ 95%.

The predicted molecular mass is 14.4 kDa per monomer and 28.9 kDa for the intact dimer (UniProt accession P04085), and the formulation is lyophilized from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid. Biological activity is determined using BALB/c 3T3 cell proliferation, with a specific activity of ≥ 2 x 10^4 units/mg and an EC50 ≤ 50 ng/mL; an example assay reported an EC50 less than 21 ng/mL after 46 hours. Endotoxin is measured at ≤ 1 EU/μg protein by kinetic LAL analysis, and cross-reactivity has been demonstrated with rat PDGF.

PDGF signals through the receptor tyrosine kinases PDGFRalpha and PDGFRbeta and acts as a potent mitogen for cells of mesenchymal origin such as fibroblasts, glial cells, and vascular smooth muscle cells. PDGF-AA mediates proliferation of oligodendrocyte progenitor cells and is commonly used to differentiate hPSC-derived neural progenitor cells into oligodendrocyte precursor cells. STEMCELL Technologies recommends storage at -20°C to -80°C; the product is stable as supplied for 12 months from the date of receipt. Reconstitute in sterile water to at least 0.1 mg/mL by pipetting down the sides of the vial without vortexing, and refer to the Safety Data Sheet, which classifies the material as not hazardous according to GHS.
