Description

A member of the interleukin-6 (IL-6) family of cytokines, Oncostatin M (OSM) bears close resemblance to leukemia inhibitory factor (LIF) and granulocyte colony-stimulating factor (G-CSF) in amino acid sequence and in its modulation of differentiation across multiple cell types. It signals through the type I receptor (gp130 and LIFR) and the type II receptor (gp130 and OSMR), activating the JAK/STAT pathway. OSM is primarily produced by activated T cells and monocytes, and also by activated macrophages, neutrophils, mast cells, and dendritic cells. Within the bone microenvironment, it is produced by cells of both hematopoietic and mesenchymal origin, including osteocytes and osteoblasts. The cytokine is involved in differentiation, cell proliferation, hematopoiesis, and inflammation, and has been implicated in liver development, bone formation, and bone resorption.

Supplied by STEMCELL Technologies, this human recombinant protein is expressed in E. coli and lyophilized after dialysis against phosphate-buffered saline. Purity is ≥ 95% as assessed by SDS-PAGE. The predicted molecular mass is 25.9 kDa (UniProt accession P13725), and endotoxin levels are ≤ 0.2 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. Biological activity is determined by a cell proliferation assay using TF-1 cells, with a specific activity of ≥ 1 x 10^5 units/mg and an EC50 ≤ 10 ng/mL. The product appears as a powder and should be reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing. Store at -80°C; the product is stable as supplied for 12 months from the date of receipt. For research use only; not intended for human or animal diagnostic or therapeutic uses.
