Description

A research-use growth factor from STEMCELL Technologies, Human Recombinant Noggin (CHO-expressed) binds and antagonizes bone morphogenetic protein (BMP) ligands of the transforming growth factor beta (TGF-β) family. It supports maintenance of undifferentiated human embryonic stem cells in vitro and can be applied short-term to prevent spontaneous differentiation. Noggin is described as essential for development of ectoderm-derived structures, embryonic somite, skeletal patterning, and neurogenesis in vivo, and as influencing chondrogenesis, osteogenesis, and joint formation, while promoting dopaminergic differentiation of embryonic stem cells and the survival of dopamine neurons.

The protein is produced in CHO cells and supplied lyophilized after dialysis against phosphate-buffered saline. Alternative names include NOG, SYM1, and SYNS1 (Accession Q13253). The predicted molecular mass is 30 kDa. Purity is ≥ 95%. Specific activity is ≥ 4 × 10^5 units/mg (EC50 ≤ 2.5 ng/mL), as determined by a bioassay using ATDC5 cells in the presence of 10 ng/mL human BMP-4. Endotoxin level is ≤ 0.2 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis. In a representative assay, the EC50 for inhibition of BMP-4 induced alkaline phosphatase production by ATDC-5 cells was 1.9 ng/mL.

The lyophilized product is stored at -80 °C and remains stable as supplied for 12 months from the date of receipt. Before use, the vial is centrifuged and reconstituted in sterile water or phosphate-buffered saline to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing. Reconstituted material should be held at 2–8 °C for up to 1 week or at -20 to -80 °C for up to 2 months, and repeated freeze-thaw cycles should be avoided.
