Hu Recom M-CSF, 100 µg

A lyophilized, E. coli-expressed human macrophage colony-stimulating factor (M-CSF) from STEMCELL Technologies is supplied as a homodimeric glycoprotein growth factor. M-CSF regulates the proliferation and differentiation of myeloid hematopoietic progenitor cells into mononuclear phagocytic lineages, including monocytes, macrophages, and osteoclasts, and is required for the maturation and activation of monocytes and macrophages. The cytokine is also involved in bone resorption by osteoclasts and contributes to the development and regulation of the placenta, mammary gland, and brain, and is produced by monocytes, fibroblasts, osteoclasts, stromal cells, endothelial cells, and tumor cells.

M-CSF exerts its biological effects by signaling through the receptor tyrosine kinase CSF-1R (also called M-CSF-R), encoded by the c-fms proto-oncogene, which activates the MAPK, PI3K, and PLCγ signaling pathways. It is used to generate monocyte-derived macrophages from immunomagnetically isolated untouched human CD14+CD16- monocytes, and can be combined with stimuli such as IFN-γ+LPS to drive M1 macrophage polarization or IL-4 for M2a polarization.

The product is lyophilized from a sterile-filtered aqueous solution containing sodium phosphate and sodium chloride at pH 8. It has a predicted molecular mass of 18.5 kDa per monomer and 37.1 kDa as the homodimer, with ≥ 95% purity. Biological activity was tested by proliferation of NFS-60 cells, with an EC50 ≤ 10 ng/mL (example range 0.9 - 1.4 ng/mL) and a specific activity of approximately 1.02 x 10^5 IU/µg, calibrated against the human recombinant M-CSF WHO International Standard (NIBSC code: 89/512). Endotoxin level is ≤ 1 EU/µg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis.
