Description

STEMCELL Technologies supplies Human Recombinant IL-8 (CXCL8), a member of the CXC subfamily of chemokines. IL-8 is a potent neutrophil chemoattractant in inflammation and activates neutrophils, inducing chemotaxis, exocytosis, and the respiratory burst. It is produced by leukocytic cells such as monocytes, T cells, neutrophils, and natural killer cells, as well as non-leukocytic somatic cells including endothelial cells, fibroblasts, and epithelial cells, with the most prominent source being monocytes and macrophages. Its production is induced by inflammatory stimuli such as IL-1. IL-8 binds to two different chemokine receptors on leukocytes, the G protein-coupled receptors CXCR1 and CXCR2, and has angiogenic effects on human intestinal microvascular endothelial cells in vitro that are mediated by CXCR2.

The biological activity of the recombinant protein was tested by its ability to mobilize Ca2+ in CHO-K1/Gα15/hCXCR1 cells (human Gα15 and human CXCR1 stably expressed in CHO-K1 cells), measured using a fluorometric assay method. The EC50, defined as the effective concentration of the growth factor at which Ca2+ mobilization is at 50% of maximum, was less than 0.15 μg/mL in the example shown. The protein has a predicted molecular mass of 8.9 kDa and was resolved with SDS-PAGE under reducing and non-reducing conditions, visualized by Coomassie Blue staining.
