Description

This animal component-free recombinant human interleukin-6 (IL-6) is supplied by STEMCELL Technologies as a lyophilized powder intended for use as a laboratory reagent. IL-6 is a pleiotropic growth factor involved in immune regulation, hematopoiesis, and oncogenesis, and is produced by a variety of cell types including T cells, B cells, monocytes and macrophages, fibroblasts, hepatocytes, vascular endothelial cells, and various tumor cell lines. On its own or in combination with factors such as IL-2 and interferon-gamma, IL-6 stimulates the proliferation of B cells, T cells, and hybridoma cells. In combination with cytokines such as IL-3, GM-CSF, and SCF, it promotes hematopoietic progenitor cell proliferation and differentiation in vitro. IL-6 also stimulates the formation of osteoclasts and affects hepatocytes, cardiomyocytes, and macrophages.

Signaling occurs through a cell surface type I cytokine receptor complex consisting of the ligand-binding IL-6 alpha (CD126) and the signal-transducing gp130 subunits, leading to activation of the JAK/STAT signaling pathway. The product is expressed in E. coli with a predicted molecular mass of 21 kDa and a stated purity of greater than or equal to 95%, lyophilized from a sterile-filtered aqueous solution containing sodium phosphate at pH 7.5. The endotoxin level is less than or equal to 1 EU/microgram of protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. Specific activity is greater than or equal to 4 x 10^7 units/mg (EC50 less than or equal to 25 pg/mL) as determined by a cell proliferation assay using B9 cells, with demonstrated cross-reactivity in mouse, rat, and monkey.

Alternative names include B cell differentiation factor, BSF-2, IFN-beta 2, and Interleukin-6, and the UniProt accession number is P05231. The vial should be stored at -20 degrees C to -80 degrees C and the product is stable as supplied for 12 months from the date of receipt. Before opening, the vial should be centrifuged and the product reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; vortexing should be avoided.
