A pleiotropic growth factor with biological activities spanning immune regulation, hematopoiesis, and oncogenesis, Interleukin-6 (IL-6) is naturally produced by T cells, B cells, monocytes and macrophages, fibroblasts, hepatocytes, vascular endothelial cells, and various tumor cell lines. The cytokine signals through a cell-surface type I cytokine receptor complex consisting of the ligand-binding IL-6Rα (CD126) and the signal-transducing gp130 subunits, inducing activation of the JAK/STAT signaling pathway.

On its own, or in combination with factors such as IL-2 and interferon-γ, IL-6 stimulates the proliferation of B cells, T cells, and hybridoma cells. In combination with cytokines such as IL-3, GM-CSF, and SCF, it has been shown to promote hematopoietic progenitor cell proliferation and differentiation in vitro. The recombinant human form of the cytokine additionally stimulates proliferation and differentiation of T cells, supports the formation of osteoclasts, and affects hepatocytes, cardiomyocytes, and macrophages.

Supplied by STEMCELL Technologies, the recombinant human IL-6 is expressed in E. coli with a predicted molecular mass of 21 kDa. It is formulated as a lyophilized powder from a sterile-filtered aqueous solution containing sodium phosphate at pH 7.5. Reported specifications include a purity of ≥ 97%, a specific activity of ≥ 4 x 10^7 units/mg (EC50 ≤ 25 pg/mL) determined by a cell proliferation assay using B9 cells, and an endotoxin level of ≤ 0.1 EU/µg protein measured by kinetic Limulus amebocyte lysate (LAL) analysis. The product is classified as not hazardous according to the Globally Harmonized System (GHS) and is intended for research use only, not for human or animal diagnostic or therapeutic applications unless otherwise stated.
