Hu Recom IL-5, ACF, 100 µg

An animal component-free recombinant human Interleukin-5 (IL-5) from STEMCELL Technologies, this cytokine is produced in E. coli and supplied as a lyophilized powder from a sterile-filtered solution containing sodium glycinate, pH 8.5. IL-5 belongs to the short-chain 4-α-helical bundle subset of hematopoietic cytokines and binds a receptor consisting of IL-5Rα and the common β-chain that is shared with the receptors for IL-3 and GM-CSF; ligand engagement triggers the JAK/STAT and MAPK signalling pathways. The cytokine is produced by Th2 cells, eosinophils, and activated mast cells, where it drives the recruitment, activation, proliferation, and survival of eosinophils and contributes to allergic inflammation, asthma, and parasite immunity. Stimulation of eosinophils with IL-5 leads to cellular activation, upregulation of CD11b expression, and inhibition of apoptosis.

The protein (UniProt accession P05113) has a predicted molecular mass of 13.3 kDa as a monomer and 26.6 kDa as a dimer, and cross-reactivity is reported to be species-specific. Purity is ≥ 95% as assessed by SDS-PAGE under reducing and non-reducing conditions with Coomassie Blue staining, and endotoxin is ≤ 1 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. Biological activity is determined by a TF-1 cell proliferation assay, with a specific activity of ≥ 4.0 x 10^6 units/mg (EC50 ≤ 0.25 ng/mL); a representative EC50 of 0.166 ng/mL is shown. The product is reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing; if not used immediately, an optional dilution in concentrated bovine serum albumin (BSA) to a final concentration of 0.1% can be prepared.

Lyophilized material is stored at -20°C to -80°C and is stable as supplied for 12 months from the date of receipt. Reconstituted stock should not be kept at 2-8°C for more than 1 month or at -80°C for more than 3 months, and repeated freeze-thaw cycles should be avoided. The Safety Data Sheet classifies the product as a non-hazardous substance and mixture, supplied as a solid. STEMCELL Technologies' quality management system is certified to ISO 13485.
