STEMCELL Technologies offers Human Recombinant IL-4 in an animal component-free formulation. Interleukin 4 plays a central role in immune regulation, driving immunoglobulin class switching in B cells (to IgE and IgG4), promoting mast cell hyperplasia and mucus production, and directing the differentiation of naïve T cells into T helper type 2 (Th2) cells. Th2 cells in turn produce IL-4, IL-5, IL-10, and IL-13.

The cytokine signals through a receptor composed of IL-4Rα and the common gamma chain, engaging JAK1/3 and recruiting STAT6 and IRS1/2. IL-4 is produced not only by Th2 cells but also by CD4+ NK T cells, γ/δ T cells, activated basophils, eosinophils, and mast cells. The human protein consists of 130 amino acids with a predicted molecular weight of 15.1 kDa, and it does not cross-react with mouse cells.

Biological activity was assessed by the ability of the recombinant IL-4 to promote proliferation of TF-1 cells, measured after 72 hours of culture using a fluorometric assay. The EC50 in this assay is 0.2 ng/mL. Protein integrity was verified by SDS-PAGE under reducing and non-reducing conditions followed by Coomassie Blue staining.
