Hu Recom IL-2, ACF, 100 µg

A research-grade recombinant human Interleukin-2 (IL-2) preparation is offered by STEMCELL Technologies in an animal component-free formulation. IL-2 is a monomeric cytokine originally identified as a T cell growth factor (TCGF; alternative name Aldesleukin; UniProt accession P60568) that binds heterotrimeric receptors consisting of CD25, CD122, and CD132, activating JAK3-, STAT5-, and AKT-dependent signaling pathways to promote cellular proliferation and survival. The cytokine is predominantly secreted by activated CD4+ and CD8+ T cells, with B cells and dendritic cells producing it in small amounts. IL-2 downregulates immune responses during thymic development, influences the development of CD4+CD25+ regulatory T cells and follicular helper T cells, and controls inflammation by inhibiting Th17 differentiation. Elevated serum IL-2 levels have been associated with progression of scleroderma, rheumatoid arthritis, and gastric and non-small cell lung cancer.

The product is expressed in E. coli and lyophilized from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid, with a predicted molecular mass of 15.5 kDa. Purity is ≥ 95%, and endotoxin level is ≤ 1 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. Biological activity is determined by a cell proliferation assay using CTLL-2 cells, with an EC50 of ≤ 3 ng/mL and a specific activity of approximately 1.8 x 10^4 IU/µg calibrated against the human recombinant IL-2 WHO International Standard (NIBSC code: 86/500). The lyophilized product is reconstituted in 10 mM hydrochloric acid to at least 0.1 mg/mL without vortexing and is stable as supplied for 12 months from the date of receipt when stored at −20°C to −80°C. The chemical is not classified as hazardous according to the GHS or the 2012 OSHA Hazard Communication Standard.
