STEMCELL Technologies supplies Human Recombinant IL-15, a four-alpha helix bundle cytokine that shares many properties with IL-2. The IL-15 receptor is a heterotrimeric complex composed of IL-15Rα (the high-affinity receptor for IL-15), IL-2/15Rβ (CD122), and the common gamma chain (CD132). IL-15 binds IL-15Rα and can be presented in trans to IL-2/15Rβ and the common gamma chain on other cells. Trans-presentation is thought to be the major mechanism of IL-15-mediated responses in mice, although it may not be required in humans. The cytoplasmic domains of IL-2/15Rβ and the common gamma chain mediate signaling that activates the JAK/STAT and PI3K pathways.

IL-15 supports the survival and proliferation of naïve CD4+ and CD8+ T cells, promotes homeostasis of memory T cells, and supports the survival and differentiation of NK cells while regulating their cytolytic activity. The protein has a predicted molecular mass of 12.9 kDa (UniProt accession P40933) and is produced in E. coli with a purity of ≥ 95%. It is lyophilized from a sterile-filtered aqueous solution containing sodium bicarbonate at pH 8.5. The endotoxin level is ≤ 1 EU/µg of protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The specific activity is ≥ 2 x 10^5 units/mg, with an EC50 of ≤ 5 ng/mL as determined by a cell proliferation assay using CTLL2 cells. Cross-reactivity has been demonstrated for mouse, rat, and monkey samples.

Store at -20°C to -80°C; the product is stable as supplied for 12 months from the date of receipt. Centrifuge the vial before opening and reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex.
