Description

STEMCELL Technologies Human Recombinant IL-12 is a heterodimeric cytokine composed of p35 and p40 subunits. It is produced by monocytes, macrophages, dendritic cells, neutrophils, and B cells in response to bacterial products and cytokines such as IFN-γ. The IL-12 receptor is expressed on T, NK, and dendritic cells, and upon binding the cytokine initiates signalling through the JAK/STAT pathway. IL-12 stimulates NK, B, and T cells to produce IFN-γ, regulates cytokine synthesis, supports the proliferation of T and NK cells, and drives the differentiation of CD4+ and CD8+ T cells.

This lyophilized recombinant protein is expressed in HEK293 cells and formulated after dialysis against phosphate-buffered saline. The predicted molecular mass of the heterodimer is 57.2 kDa, with p35 reported at 22.5 kDa (UniProt P29459) and p40 at 34.7 kDa (UniProt P29460); cross-reactivity is reported for mouse and monkey. Purity is ≥ 95%. Specific activity is ≥ 5 x 10^5 units/mg as determined by induction of IFN-γ from NK cells co-stimulated with IL-18, with an EC50 ≤ 2 ng/mL. Endotoxin level is ≤ 0.2 EU/μg protein by kinetic Limulus amebocyte lysate (LAL) analysis. According to the safety data sheet, the product is a solid powder and is not classified as a hazardous substance or mixture under the Globally Harmonized System (GHS).

Store the vial at -80°C; the product is stable as supplied for 12 months from the date of receipt. Centrifuge the vial before opening and reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial—do not vortex. As a general guideline, avoid storage at 2–8°C for more than 1 week or at -20°C for more than 2 months, and avoid repeated freeze-thaw cycles.
