Hu Recom IL-10, ACF, 1000 µg

This animal component-free recombinant cytokine is manufactured by STEMCELL Technologies using E. coli expression. It is the founding member of the IL-10 family of class II cytokines, characterized by a four-helix bundle structure of α-helical folds. Upon receptor binding, IL-10 activates signaling through JAK1 and STAT3, and is produced by dendritic cells, macrophages, CD4 T cells, mast cells, NK cells, neutrophils, and B cells under specific stimulating conditions.

The protein inhibits expression of pro-inflammatory cytokines and promotes healing processes, while enhancing B cell proliferation, immunoglobulin secretion, and class II MHC expression. IL-10 produced by macrophages also inhibits activation of neighboring macrophages, supporting self-regulation. Alternative names include BTCGF, CSIF, Cytokine synthesis inhibitory factor, Interleukin10, and TGIF.

Predicted molecular mass is 18.8 kDa, with purity ≥ 97%. The product is lyophilized from a sterile-filtered aqueous solution containing sodium phosphate at pH 7.5. Activity is determined by cell proliferation assay using MC/9 cells, with an EC50 of ≤ 5 ng/mL. Specific activity is approximately 5.4 x 10^3 IU/µg, calibrated against the human recombinant IL-10 WHO International Standard (NIBSC code: 93/722). Endotoxin level is ≤ 1 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis. Store at -20 to -80°C; the product is stable as supplied for 12 months from date of receipt. Centrifuge the vial before opening and reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. The product is not classified as hazardous under GHS and is intended for research use only.
