Description

Interleukin-10 (IL-10) is the founding member of the IL-10 family of class II cytokines, characterized by a four-helix bundle of α-helical folds. Upon receptor binding, IL-10 activates JAK1 and STAT3 signaling. It is produced by dendritic cells, macrophages, CD4 T cells, mast cells, NK cells, neutrophils, and B cells under specific stimulating conditions. IL-10 inhibits the expression of pro-inflammatory cytokines, promotes healing processes, and is important for the function of T regulatory cells. It also enhances B cell proliferation, immunoglobulin secretion, and class II MHC expression, with IL-10 produced by macrophages inhibiting activation of neighboring macrophages to provide self-regulation. STEMCELL Technologies supplies this Human Recombinant IL-10 as a laboratory reagent for research use only.

The recombinant protein is expressed in E. coli with a predicted molecular mass of 18 - 19 kDa and a purity of ≥ 95%. Biological activity is determined by a cell proliferation assay using MC/9 cells, yielding a specific activity of ≥ 5 x 10^6 units/mg (EC50 ≤ 0.2 ng/mL). Endotoxin level is ≤ 0.2 EU/μg protein as measured by kinetic limulus amebocyte lysate (LAL) analysis. The product is supplied lyophilized after dialysis against phosphate-buffered saline. According to the manufacturer's safety data sheet, it is not classified as a hazardous substance or mixture per the 2012 OSHA Hazard Communication Standard or the Globally Harmonized System (GHS), and no special protective equipment is required for normal handling. Containers should be kept tightly closed in a dry, cool, and well-ventilated place.

For reconstitution, the vial should be centrifuged before opening and the product resuspended in sterile water or phosphate-buffered saline to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; vortexing should be avoided. Reconstituted product can be stored at 2 - 8°C for up to 1 week or at -20°C to -80°C for up to 2 months, with repeated freeze-thaw cycles avoided. The lyophilized product should be stored at -80°C and remains stable as supplied for 12 months from the date of receipt.
