Description

STEMCELL Technologies supplies Human Recombinant IGF-I (Insulin-like growth factor 1) as a laboratory reagent intended for research use. IGF-I is a polypeptide belonging to the family of insulin-like growth factors and is similar in molecular structure to proinsulin. It binds to the IGF-I receptor and acts as a potent activator of the PI3K/AKT pathway while also activating ERK1/2 signaling. The factor is required for embryonic development and is produced mainly in the liver in response to a hepatocyte growth hormone. In the absence of insulin, IGF-I is necessary for the maintenance of human pluripotent stem cells.

The recombinant protein is expressed in E. coli with a predicted molecular mass of 7.7 kDa and a purity of ≥ 95%. Specific activity is ≥ 2 x 10^5 units/mg, with an EC50 ≤ 5 ng/mL as determined by a cell proliferation assay using FDC-P1 cells. Endotoxin level is ≤ 0.2 EU/µg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The material is lyophilized after dialysis against phosphate-buffered saline and should be stored at -80°C, where it is stable as supplied for 12 months from the date of receipt. Reconstitution guidance is to pipette the solution down the sides of the vial in sterile water to at least 0.1 mg/mL; the vial should be centrifuged before opening and the solution should not be vortexed.

According to the manufacturer's Safety Data Sheet, the preparation is not classified as a hazardous substance or mixture under the Globally Harmonized System (GHS) and is supplied as a solid. Cross-reactivity has been demonstrated for Mouse and Rat. The UniProt accession number is P05019.
