Description

Interferon-gamma (IFN-γ), also known as type II interferon, is a cytokine produced by T cells and NK cells, and in smaller amounts by dendritic cells and macrophages. This animal component-free human recombinant IFN-γ from STEMCELL Technologies is expressed in E. coli and supplied as a lyophilized powder from a sterile-filtered aqueous solution containing sodium phosphate, trehalose, mannitol, and TWEEN® 80 at pH 6.5. The protein has a predicted molecular mass of 16.9 kDa and a purity of ≥ 95%.

IFN-γ is controlled by cytokines such as IL-12 and IL-18 secreted in response to infection. It binds to a receptor complex and initiates signal transduction via the JAK/STAT pathway, culminating in the transcription and activation of many genes that control a diverse array of immunological functions. The cytokine stimulates the antimicrobial and anti-tumor activity of macrophages, NK cells, and neutrophils by promoting activation of microbial effector mechanisms such as the production of reactive oxygen species, nitric oxide intermediates, and complement. It also enhances the expression of major histocompatibility complex class I and II in dendritic cells and mononuclear phagocytes, favors Th1 versus Th2 cell development, and stimulates monocyte differentiation and function.

Biological activity is determined by the ability to induce cytotoxicity of HT-29 cells, with an EC50 of ≤ 0.3 ng/mL and a specific activity of ≥ 3.3 × 10⁶ units/mg. Endotoxin level, measured by kinetic Limulus amebocyte lysate (LAL) analysis, is ≤ 0.1 EU/μg protein. The product should be stored at -20 to -80°C and is stable as supplied for 12 months from date of receipt.
