Description

A lyophilized recombinant human cytokine from STEMCELL Technologies, Interferon-gamma (IFN-γ) is also known as type II interferon. It is produced by T and NK cells and, in smaller amounts, by dendritic cells and macrophages. Its expression is controlled by cytokines such as IL-12 and IL-18, and it acts by binding a receptor complex that initiates signal transduction through the JAK/STAT pathway, driving a diverse array of immunological functions including stimulation of antimicrobial and anti-tumor activity in macrophages, NK cells, and neutrophils.

The protein is expressed in E. coli and purified to > 95% purity, with a predicted molecular mass of 17 kDa (Accession Number P01579) and demonstrated cross-reactivity in mouse and monkey. Biological activity is determined in a cytotoxicity assay using HT-29 cells, yielding an EC50 of 0.035 ng/mL and a specific activity of > 2 x 10^7 units/mg (EC50 < 0.05 ng/mL). Endotoxin, measured by kinetic limulus amebocyte lysate (LAL) analysis, is < 1 EU/μg protein. The product is supplied lyophilized after dialysis against phosphate-buffered saline.

For use, the vial should be centrifuged before opening and the contents resuspended in sterile water or phosphate-buffered saline to at least 0.1 mg/mL by pipetting gently down the sides of the vial; vortexing should be avoided. The unopened product is stored at -80°C and is stable as supplied for 12 months from the date of receipt. After reconstitution, it may be kept at 2-8°C for up to 1 week or at -20°C to -80°C for up to 2 months, avoiding repeated freeze-thaw cycles. The material is classified as a laboratory reagent and is intended for research use only.
