Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a cytokine that promotes the proliferation and differentiation of hematopoietic progenitor cells and supports the generation of neutrophils, eosinophils, and macrophages. In synergy with cytokines such as stem cell factor, IL-3, erythropoietin, and thrombopoietin, it also stimulates erythroid and megakaryocyte progenitors. This Human Recombinant GM-CSF is expressed in E. coli and is supplied by STEMCELL Technologies as a lyophilized preparation. Alternative designations include colony-stimulating factor 2 (CSF-2), MGI-1GM, and Pluripoitin-alpha.

The protein has a predicted molecular mass of 14.6 kDa and is reported as not active on mouse cells. Biological activity is determined by a cell proliferation assay using TF-1 cells, with a representative EC50 of 0.08 - 0.12 ng/mL and a specific activity specification of ≥ 5 x 10^6 units/mg (EC50 ≤ 0.2 ng/mL). Purity is ≥ 95% as assessed by SDS-PAGE under reducing and non-reducing conditions, and the endotoxin level is ≤ 1 EU/μg protein by kinetic Limulus amebocyte lysate (LAL) analysis. The product is lyophilized from a sterile filtered aqueous solution containing sodium phosphate, pH 7.5, and should be stored at -20°C to -80°C, where it is stable for 12 months from the date of receipt.

GM-CSF is produced naturally by multiple cell types, including stromal cells, Paneth cells, macrophages, dendritic cells, endothelial cells, smooth muscle cells, fibroblasts, chondrocytes, and Th1 and Th17 T cells. It signals through a receptor composed of the cytokine-specific α subunit (GMRα; CD116) and the common βc subunit (CD131) shared with IL-3 and IL-5 receptors.
