Description

A recombinant human granulocyte-macrophage colony-stimulating factor (GM-CSF) from STEMCELL Technologies, supplied as an animal component-free preparation that promotes the proliferation and differentiation of hematopoietic progenitor cells and the generation of neutrophils, eosinophils, and macrophages. In synergy with cytokines such as stem cell factor, IL-3, erythropoietin, and thrombopoietin, it also stimulates erythroid and megakaryocyte progenitor cells.

The GM-CSF receptor consists of the cytokine-specific α subunit (GMRα; CD116) and the common βc subunit (CD131) shared with IL-3 and IL-5 receptors. In vivo, the recombinant protein drives the production of myeloid cells of both the granulocytic lineage (neutrophils, eosinophils, and basophils) and the monocytic lineage, and it has been investigated for mobilization of hematopoietic progenitor cells and for addressing chemotherapy-induced neutropenia in patients. It also supports the development of dendritic cells that ingest, process, and present antigens. Alternative designations include Colony-stimulating factor 2, CSF-2, MGI-1GM, and Pluripoitin-alpha.

Prepared from E. coli and supplied lyophilized in a sterile-filtered aqueous solution of sodium phosphate at pH 7.5, the protein has a predicted molecular mass of 14.6 kDa (UniProt accession P04141). Reported specifications include purity ≥ 95%, endotoxin ≤ 1 EU/μg protein by kinetic LAL analysis, specific activity ≥ 5.0 x 10^6 units/mg with an EC50 ≤ 200 pg/mL in a TF-1 cell proliferation assay, and a representative EC50 of 62 pg/mL; the product does not show activity on mouse cells. Vials should be centrifuged before opening and reconstituted in sterile water to at least 0.1 mg/mL by pipetting down the sides of the vial without vortexing; optional dilution to 0.1% BSA is suggested for working stocks. Store lyophilized material at -20°C to -80°C, where it is stable for 12 months from the date of receipt. Reconstituted stocks should not be kept at 2 - 8°C for more than 1 month or at -80°C for more than 3 months, and repeated freeze-thaw cycles should be avoided. The product is for research use only and is not intended for human or animal diagnostic or therapeutic applications.
