Description

Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a cytokine offered by STEMCELL Technologies that promotes the proliferation and differentiation of hematopoietic progenitor cells and the generation of neutrophils, eosinophils, and macrophages. In synergy with other cytokines such as stem cell factor, IL-3, erythropoietin, and thrombopoietin, it also stimulates erythroid and megakaryocyte progenitor cells. GM-CSF is produced by multiple cell types, including stromal cells, Paneth cells, macrophages, dendritic cells, endothelial cells, smooth muscle cells, fibroblasts, chondrocytes, and Th1 and Th17 T cells. It is also known by the alternative names Colony-stimulating factor 2, CSF-2, MGI-1GM, and Pluripoitin-alpha.

This animal component-free recombinant human GM-CSF is expressed in E. coli and has a predicted molecular mass of 14.6 kDa (UniProt accession P04141). Purity is ≥ 95%, and the endotoxin level is ≤ 1 EU/µg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The specific activity is ≥ 5.0 x 10^6 units/mg, with an EC50 ≤ 200 pg/mL as determined by a cell proliferation assay using TF-1 cells; a representative assay yielded an EC50 of 62 pg/mL. The product does not show activity on mouse cells and is supplied lyophilized from a sterile-filtered aqueous solution containing sodium phosphate at pH 7.5.

For preparation, the vial should be centrifuged before opening and the product reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial without vortexing. Store at -20°C to -80°C; the product is stable as supplied for 12 months from the date of receipt. According to its Safety Data Sheet, the product is classified as not a hazardous substance or mixture according to the Globally Harmonized System (GHS) and is intended for research use only.
