Glial cell line-derived neurotrophic factor (GDNF), available from STEMCELL Technologies as a human recombinant protein, is a neurotrophic factor and member of the TGF-beta superfamily. Produced in E. coli, the protein is purified to ≥ 95% purity and lyophilized from a sterile-filtered aqueous solution containing sodium citrate and sodium chloride at pH 4.0.

The recombinant factor is a homodimer composed of 15.2 kDa subunits, with a predicted total molecular mass of 30.4 kDa (UniProt accession P39905). It signals by binding the GFRα1 co-receptor and activating the RET receptor tyrosine kinase. GDNF is known to promote survival and morphological differentiation of midbrain dopaminergic neurons and to increase their high-affinity dopamine uptake. Together with BrainPhys Neuronal Medium and other supplements, it can be used to differentiate human pluripotent stem cell (hPSC)-derived neural progenitor cells into neurons.

Biological activity is determined by a C6 cell proliferation assay, with a specific activity of ≥ 3.3 x 10^2 units/mg (EC50 ≤ 3 μg/mL); an example EC50 of 2.17 μg/mL was observed. Endotoxin level is ≤ 1 EU/μg protein by kinetic LAL analysis. The lyophilized product should be stored at -20°C to -80°C and is stable as supplied for 12 months from date of receipt.
