STEMCELL Technologies supplies this animal component-free human recombinant G-CSF (granulocyte colony-stimulating factor) as a lyophilized protein expressed in E. coli. A member of the CSF family of glycoproteins, it regulates hematopoietic cell proliferation, differentiation, and function. The cytokine stimulates the initial proliferation of granulocyte-macrophage progenitors and the survival of erythroid and eosinophil progenitors, and is a key regulator of neutrophil production that drives granulocyte colony formation from hematopoietic progenitor cells. Documented actions include transient reduction of SDF-1 expression, activation of metalloproteases that cleave VCAM-1, and release of norepinephrine from the sympathetic nervous system, leading to mobilization of hematopoietic stem cells from bone marrow into the periphery. The G-CSF receptor is expressed on myeloid-committed progenitor cells, neutrophils, granulocytes, monocytes, cardiomyocytes, neuronal cells, mesothelial cells, and endothelial cells, and ligand binding activates JAK/STAT, MAPK, PI3K, and AKT signal transduction pathways.

Predicted molecular mass is 18.8 kDa and purity is ≥ 95%. Biological activity is determined by a cell proliferation assay using mouse NFS-60 cells, with an EC50 of ≤ 0.06 ng/mL (example value 0.053 ng/mL). Specific activity is approximately 5.4 x 10^4 IU/µg, calibrated against the human recombinant G-CSF WHO International Standard (NIBSC code: 09/136). The protein is reported as cross-reactive in mouse, with human as the source species (UniProt accession P09919).
