Hu Recom Flt3/Flk-2 Ligand, ACF, 100 µg

Manufactured by STEMCELL Technologies, this animal component-free recombinant cytokine is produced in E. coli and supplied as a lyophilized powder. Flt3/Flk-2 (Fms-like tyrosine kinase 3/fetal liver kinase-2) Ligand is a hematopoietic cytokine that acts as a co-stimulatory factor in the proliferation, differentiation, and survival of hematopoietic stem and progenitor cells and supports the development of the immune system. The ligand exists as both membrane-bound and soluble isoforms, with both being biologically active, and signals through the class III tyrosine kinase receptor Flt3/Flk-2 (CD135).

Together with stem cell factor and thrombopoietin, the product is commonly used to expand primitive CD34+ hematopoietic cells in culture. In combination with myeloid cytokines such as GM-CSF, G-CSF, or M-CSF, it enhances the growth of clonogenic myeloid progenitor cells, and in synergy with IL-3, IL-4, IL-7, IL-11, IL-12, IL-15, GM-CSF, and TNF-α it regulates the development of lymphoid progenitor cells, including dendritic cell, B cell, T cell, and NK cell progenitors. It has no significant effect on erythropoiesis or megakaryopoiesis.

The protein has a predicted molecular mass of 17.6 kDa and is ≥ 95% pure. Specific activity is ≥ 1 x 10^5 units/mg, with an EC50 ≤ 10 ng/mL as determined by cell proliferation using OCI-AML5 cells. Endotoxin level is ≤ 1 EU/μg protein as measured by kinetic LAL analysis, and cross-reactivity is observed with mouse, rat, and monkey. The product is lyophilized from a sterile-filtered solution containing sodium phosphate and sodium chloride at pH 7.5 and should be stored at -20°C to -80°C, where it remains stable for 12 months from date of receipt. The vial should be centrifuged before opening and reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial, without vortexing.
