STEMCELL Technologies supplies this Human Recombinant FGF-10 (KGF-2) as a laboratory reagent intended for research use. The recombinant protein is produced in E. coli and presented as a lyophilized powder prepared by dialysis against phosphate-buffered saline. Purity is ≥ 95% and the endotoxin level is ≤ 0.2 EU/μg protein, as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The product is provided as a solid with no reported odor.

The protein has a predicted molecular mass of 19.3 kDa and binds with high affinity to fibroblast growth factor receptor 2 IIIb (FGFR2 IIIb), with weaker affinity for FGFR1 IIIb. Its specific activity is ≥ 5 x 10^4 units/mg, with an EC50 of ≤ 20 ng/mL determined using a fluorometric cell proliferation assay with 4MBr-5 cells. FGF-10 mediates the epithelial–mesenchymal interactions essential to lung development, and has been described as supporting mobilization and proliferation of lung-resident mesenchymal stem cells (MSCs), protection and repair in models of acute lung injury, and endodermal differentiation of human pluripotent stem cells toward insulin-producing pancreatic-like cells. Cross-reactivity has been demonstrated with mouse and rat.

For storage, the product is held at -80°C and is reported stable as supplied for 12 months from the date of receipt. The vial should be centrifuged before opening and reconstituted in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; vortexing should be avoided.
