Fibroblast growth factor 10 (FGF-10) is a member of the fibroblast growth factor family predominantly expressed by mesenchymal fibroblasts during embryonic development. It binds with high affinity to fibroblast growth factor receptor 2 IIIb (FGFR2 IIIb) and has a weaker affinity for FGFR1 IIIb. FGF-10 and FGF-7 share similar receptor binding properties and target cell specificities but are differentially regulated by components of the extracellular matrix.

This Human Recombinant FGF-10 (KGF-2) from STEMCELL Technologies mediates epithelial-mesenchymal interactions essential to lung development. It also plays a role in mobilization and proliferation of lung-resident mesenchymal stem cells (MSCs), protection and repair against acute lung injury, and endodermal differentiation of human pluripotent stem cells to insulin-producing pancreatic-like cells. The protein is produced in E. coli and purified to ≥ 95%, with a predicted molecular mass of 19.3 kDa and cross-reactivity against mouse and rat. Biological activity is determined using a cell proliferation assay with 4MBr-5 cells measured by a fluorometric assay method, yielding an EC50 of less than 20 ng/mL and a specific activity of ≥ 5 x 10^4 units/mg. Endotoxin level is ≤ 0.2 EU/μg protein, measured by kinetic Limulus amebocyte lysate (LAL) analysis.

The product is supplied lyophilized after dialysis against phosphate-buffered saline. Reconstitute in sterile water to at least 0.1 mg/mL; centrifuge the vial before opening and do not vortex. Store at -80°C; the product remains stable as supplied for 12 months from the date of receipt. As a general guide, do not store at 2–8°C for more than 1 week or at -20°C for more than 3 months, and avoid repeated freeze-thaw cycles.
