Description

Erythropoietin (EPO) is a glycoprotein growth factor that is produced primarily in the kidney in response to hypoxia or anemia. It is the principal physiological regulator of erythropoiesis. EPO promotes erythropoiesis by binding to a homodimeric cell surface receptor that activates JAK2/STAT5, PI3K/AKT, and MAPK pathways, and stimulates the proliferation and differentiation of erythroid progenitor cells. From STEMCELL Technologies, this Human Recombinant EPO is expressed in CHO cells and is also known as Epoetin and Erythropoietin (UniProt accession P01588).

The recombinant polypeptide has a predicted molecular mass of 18.4 kDa, and as a result of glycosylation it migrates with an apparent molecular mass of 26 – 36 kDa in SDS-PAGE. Cross-reactivity is reported with mouse and rat. The product is supplied lyophilized after dialysis against phosphate-buffered saline (PBS).

Activity is determined by a cell proliferation assay using TF-1 cells, with an EC50 ≤ 0.6 ng/mL. Specific activity is approximately 220 IU/μg as determined by titration in a CFU assay on human bone marrow MNCs, calibrated against the third international reference preparation of EPO (NIBSC code: 11/170). Purity is ≥ 95%, and endotoxin level is ≤ 0.2 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. The vial should be stored at -80°C and is stable for 12 months from date of receipt. Before opening, centrifuge the vial, then reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. Stock solutions may be stored at 2 – 8°C for up to 1 week or at -20°C for up to 2 months, and repeated freeze-thaw cycles should be avoided.
