Description

A glycoprotein growth factor produced primarily in the kidney in response to hypoxia or anemia, erythropoietin (EPO) from STEMCELL Technologies is the principal physiological regulator of erythropoiesis. The human recombinant protein is expressed in CHO cells and promotes erythropoiesis by binding to a homodimeric cell surface receptor that activates JAK2/STAT5, PI3K/AKT, and MAPK pathways, stimulating the proliferation and differentiation of erythroid progenitor cells.

Alternative names include Epoetin and Erythropoietin, with accession number P01588. The polypeptide has a predicted molecular mass of 18.4 kDa and, due to glycosylation, migrates with an apparent molecular mass of 26 - 36 kDa in SDS-PAGE. Cross-reactivity has been reported with mouse and rat. The product is lyophilized after dialysis against phosphate-buffered saline (PBS), with a purity of ≥ 95% and an endotoxin level of ≤ 0.2 EU/μg protein measured by kinetic Limulus amebocyte lysate (LAL) analysis.

Biological activity is confirmed in TF-1 cell proliferation assays, with an EC50 ≤ 0.6 ng/mL and a specific activity of approximately 220 IU/μg, calibrated against the third international reference preparation of EPO (NIBSC code: 11/170). Activity is also validated by titration in a CFU assay on human bone marrow mononuclear cells, with colonies enumerated using STEMvision™. The product should be stored at -80°C, where it remains stable for 12 months from date of receipt, and reconstituted in sterile water to at least 0.1 mg/mL before use.
