Description

Supplied by STEMCELL Technologies, this Bone Morphogenetic Protein 4 (BMP-4) is a recombinant human protein belonging to the highly conserved transforming growth factor β (TGF-β) superfamily. The mature protein is a disulfide-linked homodimer consisting of two 116-amino-acid residue subunits, generated by proteolytic removal of the signal peptide and propeptides.

BMP-4 binds to type I and type II receptors on cells, triggering phosphorylation of receptor 1 and subsequently of Smad proteins, which then act as transcription factors. BMP-4 has been identified as a key regulator of embryogenesis and is known to play a role in the growth and differentiation of various cell types, including embryonic stem cells (ESCs), induced pluripotent stem cells (iPSCs), mesenchymal cells, epithelial cells, hematopoietic cells, and neuronal cells. Alternative names include BMP-2B, BMP2B1, MCOPS6, OFC11, and ZYME (UniProt accession P12644).

Manufactured in CHO cells and supplied lyophilized from a sterile-filtered solution containing 0.1% trifluoroacetic acid, the protein has a predicted molecular mass of 13.1 kDa (monomer) or 26.2 kDa (dimer), migrating on SDS-PAGE to an apparent molecular mass of 20 kDa or 34 kDa, respectively, due to variable glycosylation. Purity is ≥ 95%. Specific activity is ≥ 6.7 x 10^4 units/mg, with an EC50 ≤ 15 ng/mL, as determined by alkaline phosphatase activity induced in ATDC-5 cells; an example EC50 is 3.99 ng/mL. Endotoxin level is ≤ 1 EU/μg protein. Store at -20 to -80 °C; stable as supplied for 12 months from date of receipt. For research use only.
