Description

Manufactured by STEMCELL Technologies, this recombinant Brain-Derived Neurotrophic Factor (BDNF) is a member of the NGF family of neurotrophins that supports the differentiation and survival of specific neuronal subpopulations in both the central and peripheral nervous systems. The protein binds with high affinity to the TrkB receptor and activates AKT and ERK signaling pathways. It is expressed in the hippocampus, cortex, and synapses of the basal forebrain, and acts as a survival factor for human embryonic stem cells when plated on feeder cells or Corning® Matrigel®. The product also exhibits cross-reactivity with mouse and rat BDNF.

BDNF regulates synaptic transmission and plasticity at adult synapses, contributing to long-term potentiation, long-term depression, certain forms of short-term synaptic plasticity, and homeostatic regulation of neuronal excitability. It also promotes survival and growth of dorsal root ganglion cells and hippocampal and cortical neurons. In combination with GDNF and other supplements, BDNF can be used to differentiate hPSC-derived neural progenitor cells into neurons. The protein is produced in E. coli and supplied as a homodimer of 13.6 kDa subunits with a predicted total molecular mass of 27.3 kDa. Alternative names include Abrineurin, ANON2, BULN2, MGC34632, and Neurotrophin (UniProt accession P23560).

The product is lyophilized from a sterile-filtered solution containing 0.1% trifluoroacetic acid, with a purity of ≥ 95% and an endotoxin level of ≤ 1 EU/μg protein by kinetic LAL analysis. For lot #1000038506 or higher, specific activity is ≥ 2.5 x 10^4 units/mg (EC50 ≤ 40 ng/mL) using a TrkB-expressing neuroblastoma cell line (BR6); for all other lots, specific activity is ≥ 5.0 x 10^2 units/mg (EC50 ≤ 2 μg/mL) using C6 cells. The lyophilized product should be stored at -20°C to -80°C and is stable for 12 months from the date of receipt. Reconstitution in sterile water to at least 0.1 mg/mL is recommended; if the reconstituted product will not be used immediately, optional dilution with concentrated bovine serum albumin to a final concentration of 0.1% is suggested, and repeated freeze-thaw cycles should be avoided. For research use only.
