Hu Recom BDNF, 100 µg

A member of the NGF family of neurotrophins, this recombinant Brain-derived neurotrophic factor (BDNF) from STEMCELL Technologies is required for the differentiation and survival of specific neuronal subpopulations in both the central and peripheral nervous systems. The human protein binds with high affinity to the tropomyosin receptor kinase B (TrkB) and activates the AKT and ERK pathways. It is expressed in the hippocampus, cortex, and synapses of the basal forebrain, where it regulates synaptic transmission and plasticity at adult synapses in the central nervous system.

BDNF contributes to adaptive neuronal processes such as long-term potentiation, long-term depression, certain forms of short-term synaptic plasticity, and homeostatic regulation of neuronal excitability, and promotes survival and growth of dorsal root ganglion cells and hippocampal and cortical neurons. It also acts as a survival factor for human embryonic stem cells plated on feeder cells or Corning® Matrigel®, and together with glial cell line-derived neurotrophic factor (GDNF) and other supplements is commonly used to differentiate human pluripotent stem cell (hPSC)-derived neural progenitor cells into neurons. The protein demonstrates cross-reactivity with mouse and rat.

The product is supplied lyophilized from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid and is expressed in E. coli. The predicted molecular mass is 13.6 kDa for the monomer and 27.3 kDa for the dimer, with purity ≥ 95% and endotoxin level ≤ 1 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. Alternative names include Abrineurin, ANON2, BULN2, MGC34632, and Neurotrophin (UniProt accession P23560). For Lot #1000038506 or higher, specific activity is ≥ 2.5 × 10^4 units/mg (EC50 ≤ 40 ng/mL) by cell proliferation assay using a neuroblastoma cell line stably expressing TrkB (BR6); for other lots, specific activity is ≥ 5.0 × 10^2 units/mg (EC50 ≤ 2 μg/mL) by cell proliferation assay using C6 cells. Store at -20°C to -80°C; stable as supplied for 12 months from date of receipt. Reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. For research use only.
