Description

Growth Differentiation Factor 11 (GDF-11), a member of the transforming growth factor beta (TGF-β) family, is available from STEMCELL Technologies in an animal component-free recombinant form covering both human and mouse sequences. GDF-11 binds to the TGF-β receptors ALK4, ALK5, and ALK7 and activates the SMAD signaling pathway. It plays roles in the development of the olfactory system, retina, and pancreas, as well as in anterior/posterior patterning of the axial skeleton. The protein also functions as an endocrine factor expressed in skeletal muscle, brain, and dental pulp. Studies in mice have shown that GDF-11 regulates muscle and cardiac aging and stimulates neurogenesis by remodeling blood vessels.

The recombinant protein is produced in E. coli and is supplied lyophilized from a sterile-filtered solution containing 0.1% trifluoroacetic acid. It has a predicted molecular mass of 24.9 kDa (12.5 kDa per monomer). Specific activity is ≥ 1.0 × 10^4 units/mg as determined by an alkaline phosphatase activity assay using ATDC-5 cells, with an EC50 ≤ 100 ng/mL. Purity is ≥ 95%, and endotoxin level is ≤ 1 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis.

For storage, the product should be kept at -20°C to -80°C and remains stable as supplied for 12 months from the date of receipt. Centrifuge the vial before opening, then reconstitute in 10 mM hydrochloric acid to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex. The product is classified as not a hazardous substance or mixture according to the Globally Harmonized System (GHS) and is intended for research use only.
