Description

A recombinant bone morphogenetic protein 2 (BMP-2) from STEMCELL Technologies, supplied as an animal component-free preparation. BMP-2 is a member of the transforming growth factor beta (TGF-β) superfamily and forms a disulfide-linked homodimer that acts as a ligand for complexes of type I and II BMP receptors, primarily activating SMAD1/5/8 signaling. It is a potent differentiation factor that directs human pluripotent stem cells toward extra-embryonic endoderm, mesenchymal cells, and chondrocytes, induces bone and cartilage formation in vitro, and promotes chondrogenesis in human mesenchymal stem cells. While BMP-2 expression is low in healthy cartilage, expression is upregulated at sites of cartilage damage.

The protein is expressed in E. coli and provided as a lyophilized solid from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid. The predicted molecular mass is 13 kDa for the monomer and 26.1 kDa for the dimer. Specifications include ≥ 95% purity and ≤ 1 EU/μg endotoxin by kinetic Limulus amebocyte lysate (LAL) analysis. Specific activity is ≥ 4.0 x 10^3 units/mg, with an EC50 ≤ 250 ng/mL as determined by alkaline phosphatase activity induced in ATDC-5 cells; a representative assay produced an EC50 of 74 ng/mL.

Reconstitute in sterile water to at least 0.1 mg/mL by pipetting down the sides of the vial, without vortexing. Store at -20°C to -80°C; the product is stable as supplied for 12 months from date of receipt. For research use only.
