Hu/Ms Recom Activin A, ACF, 100 µg

From STEMCELL Technologies, this animal component-free preparation of Activin A is a disulfide-linked homodimer composed of two beta-A chains and a member of the transforming growth factor beta (TGF-β) family. Active in both human and mouse systems, the protein binds to heteromeric complexes of a type I (Act RI-A and Act RI-B) and a type II (Act RII-A and Act RII-B) serine-threonine kinase receptor, and signals through SMAD2/3 proteins. It regulates a variety of functions, including cell proliferation, differentiation, wound healing, apoptosis, and metabolism, maintains the undifferentiated state of human embryonic stem cells, and facilitates differentiation of human embryonic stem cells into definitive endoderm.

Produced in E. coli and supplied as a lyophilized powder, the product is formulated from a sterile-filtered aqueous solution containing 0.1% trifluoroacetic acid. Specifications include a predicted molecular mass of 13.1 kDa for the monomer and 26.2 kDa for the dimer, a purity of ≥ 95%, and an endotoxin level of ≤ 1 EU/μg protein as measured by kinetic Limulus amebocyte lysate (LAL) analysis. Biological activity is determined by a cell proliferation assay using MPC-11 cells, with an EC50 of ≤ 10 ng/mL; in a representative assay the EC50 was 0.436 ng/mL. The specific activity is approximately 1.2 IU/µg as calibrated against the human recombinant Activin A WHO International Standard (NIBSC code: 91/626).

Centrifuge the vial before opening and reconstitute in sterile water to at least 0.1 mg/mL by pipetting the solution down the sides of the vial; do not vortex.
